One of the main characters in the game is the military brother of Bottles, Jamjars. This name was actually first used in Banjo-Kazooie during the opening training. As the player is testing out Kazooie's beak barge, Bottles comments that Kazooie is useful for something, to which she retaliates with, "I'm trying this move out on you next, jam jars!" The term comes from slang for large glasses with thick lenses.

There is evidence in the code of the arcade version that the two boss characters, Huitzil and Pyron, may have been planned as playable at one stage of development. There is an alternate color scheme for both of the characters that shows up if you are playing a mirror match, both characters are able to be played in Arcade mode though hacking (though Pyron can't fight the final boss due to a bug), both of the characters have their own endings, which are the exact same as their endings in the sequel, "Night Warriors: Darkstalkers' Revenge", though in a more unfinished state with the ending text just repeating "A" and the music not playing like it should.

In a 1996 interview published in a Dungeons & Dragons mook (magazine & book portmanteau) covering Dungeons & Dragons: Shadow over Mystra and this game, the developers were asked why they choose Dungeons & Dragons as the setting. The game's designer Tomoshi Sadamoto responded: "It was TSR, the copyright holder of D&D, who came to us first and talked about doing a game. However, an agreement could not be reached at that point and the project was on hold for awhile. Sometime later a compromise was reached, and we were able to start working on the first game, Tower of Doom." He also admitted that from the beginning, the game was only intended for the arcade.

Sony's decision to bring Vib-Ribbon to the PlayStation Network came as a result of Sony Computer Entertainment of America's President and CEO, Shawn Layden, throwing out the name on-stage at E3 2014, unaware that Vib-Ribbon was never originally released in North America. The outcry from fans to release it in North America following the conference spurred Layden to demand a port of it for American consoles.
